You have to defeat all of the Big Heads. Then, you have to open the south-east area using the bone numbers.

P.S. The area I'm talking about is roughly in the center of the Tomb of Bones.

Just go right from the entrance, fight off an ambush, cross the bridge, and you'll enter a room with bones in it. You have to arrange the bones in certain patterns to open the rooms where the Big Heads are. There are three of them, Todd, Rod, and Maude, and they all have different attacks.
